Antioxidant
Pterostilbene has been shown to help reduce free radical damage. When compared to tetrahydrocurcumin, photoglycopyrrolidine, and vitamin C, astragalus has the strongest oxygen radical uptake; and has a strong ability to inhibit DPPH free radicals, which suggests a superb antioxidant effect. In published studies, pterostilbene has been found to display direct antioxidant effects while also modulating antioxidant enzymes and defense systems throughout the body. No known botanical ingredient has surpassed this to date.
Inhibition of Melanin
Pterostilbene is a unique autophagy inducer, that can effectively induce cytoprotective autophagy in human umbilical vein endothelial cells by rapidly increasing intracellular calcium concentration and AMPKa1 activation, and thus inhibiting mTOR, a negative regulator of autophagy. Pterostilbene plays an inhibitory role against melanogenesis by down-regulating proteins involved in α-MSH-triggered B16-F10 cells and induces resistance through autophagy to melanogenesis.
Spot Removal
In the assay experiments for the efficacy of spot removal, the untreated cells had obvious black spots; while the cells were treated with 0.55 μg/ml of pterostilbenes (90%), used every 3 days for 9 days, and eventually the black spots disappeared, indicating that it has a good effect of spot removal.
Whitening
Pterostilbene has excellent melanin and complexin inhibiting effects. Studies have shown that it inhibits melanogenesis, melanocyte dendrites and melanosome transport via the cAMP/PKA/CREB pathway. Its half inhibitory concentration IC50 value for tyrosinase is significantly better than that of common whitening ingredients such as vitamin C and arbutin, and only slightly lower than that of photoglycoside. When compared with whitening ingredients such as glycyrrhizin, tretinoin and vitamin C, pterostilbene has the most significant melanin inhibiting effect. Not only that, pterostilbene can also promote cell autophagy and effectively metabolize melanin and lipofuscin that have already been generated in the skin, so its whitening speed is more rapid than that of common whitening ingredients.
Protects against UV Damage
Studies have shown that pterostilbene can express antioxidant enzymes and has good UVB protective efficacy. In cellular experiments, 30 μg/mL of pterostilbene can reduce half of the cytotoxic damage caused by UVB; and a low concentration of 4 μmol/L of pterostilbene pretreatment can inhibit the DNA damage of skin cells by UVA irradiation. In addition, the use of the sun protection factor (SPF value) test in human experiments revealed that the SPF value of the cream containing 0.4% pterostilbene was 6.2, which demonstrated the effect of pterostilbene as a natural sunscreen.
Anti-Inflammatory
Through the regulation of the Nrf2 pathway, pterostilbene can effectively inhibit the expression of inflammatory factors such as COX-2, PGE2, and NO, demonstrating excellent anti-inflammatory effects. Meanwhile, pterostilbene is also able to reduce intracellular inflammatory responses and reactive oxygen species (ROS) levels and inhibit neutrophil activation, inflammatory mediator production, and endothelial cell apoptosis.
